Goodwin BL, Pendleton LC, Levy MM, Solomonson LP, Eichler DC. Tumor necrosis factorreduces argininosuccinate synthase expression and nitric oxide production in aortic endothelial cells. Am J Physiol Heart Circ Physiol 293: H1115–H1121, 2007. First published May 11, 2007; doi:10.1152/ajpheart.01100.2006.—Endothelial dysfunction associated with elevated serum levels of TNFobserved in diabetes, obesity, and congenital heart disease results, in part, from the impaired production of endothelial nitric oxide (NO). Cellular NO production depends absolutely on the availability of arginine, substrate of endothelial nitric oxide synthase (eNOS). In this report, evidence is provided demonstrating that treatment with TNF(10 ng/ml) suppresses not only eNOS expression but also the availability of arginine via the coordinate suppression of argininosuccinate synthase (AS) expression in aortic endothelial cells. Western blot and real-time RT-PCR demonstrated a significant and dose-dependent reduction of AS protein and mRNA when treated with TNFwith a corresponding decrease in NO production. Reporter gene analysis demonstrated that TNFsuppresses the AS proximal promoter, and EMSA analysis showed reduced binding to three essential Sp1 elements. Inhibitor studies suggested that the repression of AS expression by TNFmay be mediated, in part, via the NFB signaling pathway. These findings demonstrate that TNFcoordinately downregulates eNOS and AS expression, resulting in a severely impaired citrulline-NO cycle. The downregulation of AS by TNFis an added insult to endothelial function because of its important role in NO production and in endothelial viability.

Endothelial argininosuccinate synthetase 1 regulates nitric oxide production and monocyte adhesion under static and laminar shear stress conditions.

Laminar shear stress (LSS) is known to increase endothelial nitric oxide (NO) production, which is essential for vascular health, through expression and activation of nitric oxide synthase 3 (NOS3). Recent studies demonstrated that LSS also increases the expression of argininosuccinate synthetase 1 (ASS1) that regulates the provision of L-arginine, the substrate of NOS3. It was thus hypothesize...

EXPRESSION OF INDUCIBLE NITRIC OXIDE SYNTHASE GENE (iNOS) STIMULATED BY HYDROGEN PEROXIDE IN HUMAN ENDOTHELIAL CELLS

Inducible nitric oxide synthase (iNOS) gene expresses a calcium calmudolin-independent enzyme which can catalyse NO production from L-arginine. The induction of iNOS activity has been demonstrated in a wide variety of cell types under stimulation with cytokines and lipopoly saccharide (LPS). Previous studies indicated that all nitric oxide synthases (NOS) activated in human umbilical vein endot...

Co-regulation of AS and eNOS phosphorylation by calcium 1 Protein Kinase C α Phosphorylates a Novel Argininosuccinate Synthase Site at Serine 328 During Calcium-Dependent Stimulation of Endothelial Nitric Oxide Synthase in Vascular Endothelial Cells*

Endothelial nitric oxide synthase (eNOS) utilizes L-arginine as its principal substrate, converting it to L-citrulline and nitric oxide (NO). L-citrulline is recycled to L-arginine by two enzymes, argininosuccinate synthase (AS) and argininosuccinate lyase (AL), providing the substrate arginine for eNOS and NO production in endothelial cells. Together, these three enzymes, eNOS, AS and AL, make...

Induction of endothelial nitric oxide synthase expression by melanoma sensitizes endothelial cells to tumor necrosis factor-driven cytotoxicity.

PURPOSE The cascade of molecular events leading to tumor necrosis factor (TNF)-mediated tumor regression is still incompletely elucidated. We investigated the role of endothelial nitric oxide synthase in determining the tumor-selective activity of TNF. EXPERIMENTAL DESIGN Using quantitative real-time PCR, endothelial nitric oxide synthase gene levels were measured in melanoma metastases of th...
